No, your sibling can not apply for a U.S. tourist visa WHILE he/she is also a Tourist visa holder in Japan. Your sibling must go back and apply to her/his point of origin because she/he has no strong ties with Japan.
just to give you an idea:

(日本語) (http://japan.usembassy.gov/j/visa/tvisaj-niv-b2.html) Frequently Asked Questions (http://japan.usembassy.gov/e/visa/tvisa-niv-b1b2faq.html)
Travellers wishing to visit the United States for pleasure may apply for a temporary visa. The visa symbol is "B-2".
The holder of a B-2 visa may undertake any one of the following activities:
Tourism
Visit friend/relatives
Undergo medical treatment
Participate in conventions, conferences or convocations of fraternal or social organizations
Amateur participation in musicals, sports, or similar eventsTo obtain a B-2 visa, you must show that:
You have strong ties to a residence in foreign country to which you do not intend to abandon
Your trip to the United States will be for a definite temporary period and that you will return upon the conclusion of your visit
You have sufficient funds to cover the expenses of the visit and return passage.
There is no specific list of documents which must be submitted to prove each of these points.
As an example, however, to show residence in Japan, you must show that you have ties to this country. Proof of employment, family ties or other connections, or home ownership will often satisfy this requirement. Substantiating documents should be submitted with your application. Old passports containing previous visas are also useful.

Normally di hinihingi certificate of bank deposit or any other proof of an existing account kasi di naman nakalagay as requirements na kelangan i-present. See this link as reference, http://japan.usembassy.gov/e/visa/tvisa-nivapply.html
Pero depende kung mahigpit ang mag-iinterview sayo baka humingi rin sila nyan as additional requirements kasi walang specified docs na nasa category na yan.:)

in my case, hinanapan ako
100 lapad po ang pinakita ko (time deposit)
awa naman ng Diyos nabigyan ako ng 10 yrs multiple
to think na 1month lang ang hinihingi kong visa that time
kala ko nga typhographical error

Case by case talaga, na-mentioned ko sa ibang thread sabay kami ng friend ko nag-apply at ako nag-fill up ng application namin online. Kaibahan lang pinalagay ng friend ko na may bibisitahin syang relative sa US at ako naman tour lang with family. Na-deny sya at ako nakakuha then re-apply sya at ginaya reason ko to visit nabigyan din ng 10yrs multiple.:)
